PBMCs from a Healthy Donor: Paired T Cell Receptor

Human peripheral blood mononuclear cells (PBMCs) of a healthy female donor aged 29 were obtained by 10x Genomics from AllCells. The whole transcriptome (parent) library was prepared with Chromium Single Cell V(D)J Reagent Kits User Guide (v1 Chemistry) with Feature Barcode technology for Cell Surface Protein (CG000186), and using the Chromium Single Cell V(D)J Enrichment Kit Human T Cell (PN-1000005) and sequenced on an Illumina NovaSeq 6000.

Single Cell Paired TCR Library

  • Sequencing Depth: 14,125 read pairs per cell
  • Paired-end (150 x 150), single indexed Read 1: 150 cycles (16 bp barcode, 10 bp UMI) i7 index: 8 cycles Read 2: 150 cycles

The key metrics were:

  • Estimated number of cells: 3,981
  • Cells with Productive V-J Spanning Pair: 3,364 cells
  • Reads mapped to any VDJ gene: 72.9%
  • Cells detected contain a productive V-J spanning receptor pair: 84.5%
