Nuclei were isolated from ~25 mg of fresh frozen C57/Bl6 adult mouse brain purchased from BioIVT following the Chromium Nuclei Isolation Reagent Kits Sample Prep User Guide (CG000505).
Gene Expression libraries were generated as described in the Chromium Single Cell 3' Reagent Kits User Guide (v3.1 - Dual Index) User Guide (CG000315) and sequenced on an Illumina NovaSeq 6000 with approximately 60,000 read pairs per cell.

- 90 cycles Read 2
The cellranger multi pipeline in Cell Ranger v8.0.0 was used to analyze libraries.
Note: The 'High Fraction of Reads Mapped Antisense to Genes' alert in the web summary is expected because this is a nuclei sample. The value (~24%) is in line with historical data for nuclei using SC3Pv3 chemistry.
