Peripheral blood mononuclear cells (PBMCs) from a C57BL/6 mouse strain were obtained by 10x Genomics from iQ Biosciences. Splenocytes from a C57BL/6 mouse strain were obtained by 10x Genomics from iQ Biosciences.
Gene Expression and Cell Surface Protein libraries were generated as described in the GEM-X Universal 5' Gene Expression v3 4-plex User Guide (CG000770) and sequenced on an Illumina NovaSeq 6000 with approximately 25,000 read pairs per cell for the Gene Expression library, 33,000 read pairs per cell for the VDJ-T library and 15,000 read pairs per cell for the VDJ-B library.
Paired-end, dual indexing libraries were sequenced with the following configuration:
- 28 cycles for Read 1
- 10 cycles for i7
- 10 cycles for i5
- 90 cycles for Read 2
Libraries were analyzed using the cellranger multi pipeline.
