Single cell gene expression offers crucial insights into the identity and function of single cells. However, there is much to be understood about the regulatory landscape that controls gene expression. In order to see how chromatin compaction and DNA-binding proteins regulate gene expression, scientists need to be able to profile the chromatin of tens of thousands of single cells in parallel. With the resolution of single cells, researchers can further decipher heterogeneous cell types and states by unmasking epigenetic differences. This is where the Chromium Single Cell ATAC (Assay for Transposase Accessible Chromatin) Solution comes in.

Named one of the Top 10 Innovations of 2019 by The Scientist, the Chromium Single Cell ATAC Solution leverages a transposase enzyme to tag accessible regions of chromatin with adaptors. These regions are sequenced, and can be used to infer and map transcription factor binding sites, among other regulatory structures, and to build gene regulatory networks.

Already, single cell ATAC-seq is helping to address major questions in the study of cancer. Researchers from Stanford University applied single cell ATAC-seq to study T cell exhaustion in cancer. They characterized the chromatin regulators of therapy-responsive intratumoral T cells after PD-1 blockade in patients with basal cell carcinoma, helping to resolve some of the complex biology underlying the variable patient response to immunotherapies.
